Typewriter Therapeutics is developing an all-RNA gene-writing platform that inserts therapeutic genes at a defined genomic site without viral vectors or double-strand DNA breaks. Its programs target in vivo engineered CAR-T cells and gene insertion for severe genetic diseases, including liver disorders.
Typewriter's platform uses the R2 retrotransposon system to insert a therapeutic gene at a natural, defined genomic safe-harbor site. The two-part approach delivers RNA in lipid nanoparticles; the R2 machinery binds its RNA, nicks one DNA strand and uses that site to reverse-transcribe the RNA into DNA. The company says the method avoids viral vectors and double-strand breaks while aiming for gene expression that persists as cells divide.
The company is pursuing two applications: engineering CAR-T cells in vivo and inserting genes into liver cells for severe rare diseases. Typewriter is in platform and program development, with its Series A supporting advancement of these genetic-medicine programs; these are investigational approaches rather than approved therapies.
