Developing elismetrep through clinical trials

TriGemX Bio, formerly Kallyope, is developing elismetrep, an oral drug candidate intended to treat a migraine attack. It blocks TRPM8, an ion channel involved in neural signaling that human genetic studies have linked to migraine risk. This target differs from the CGRP pathway addressed by an existing class of migraine medicines. The intended beneficiaries are people whose attacks are not adequately controlled by available therapies; this is a medicine in development rather than a treatment currently sold to patients.

The company has advanced elismetrep through a nearly 450-patient Phase 2b study and into Phase 3. Its current program includes a pivotal trial and a long-term safety study, with a second pivotal trial scheduled to begin in October 2026. Initial Phase 3 results are expected in mid-2027. Elismetrep has not been approved by the FDA or another regulatory agency; those studies are intended to establish the evidence for a potential future approval.
